Can you actually run from sunspear great hall to desolation, and therefore skipping all the content in between?

No, because you need to do the quests in order to unlock the quest,

Crossing the Desolation

There is no other way past it.

The only way to enter the Realm of Torment is by doing the Ruins of Morah mission to open the Vortex, and this mission is started by talking to an NPC in the outpost. Mission NPCs in Nightfall will not start a mission unless everyone in the party has done all primary quests and missions leading up to that mission. So, even if you could somehow get a run as far as Ruins of Morah, it wouldn't put you any closer to DoA.

you can get ferry to the docks at early level but i dont know if you can get into the mission without the main quest, but the rest of the game you must follow the story line

You can enter into the Domain of Anguish only if you have beaeten Abaddon, aka finished Nightfall. And to get to Abaddon's Gate you have to do Ruins of Morah, as others mentioned.

Oh, and of course you would definately need Chantry of Secrets unlocked :P

Obviously not. Unless you can swim, you'll need to do the Consulate Docks mission to get off the island at least.

(If you meant the Sunspear Sanctuary instead, the answer is still no.)
